Paired-end shotgun sequencing

Paired-end shotgun sequencing works by first creating multiple copies of the human genome. Then, each copy of the genome is “shotgunned” into smaller fragments, with the breakages occurring at random points in the genome. The ends of the fragments are sequenced and a computer algorithm matches overlapping ends together to piece the genome back together.
